The obvious first choice:

Peter Parker/Spider-Man: Titular Web-Spinning, Wall-Crawling, Wise-Cracking Arachnid-Themed Defender of New York. At hand to take on and take down criminals and supervillians who lay beyond the reach of standard Law Enforcement.

Dr Curtis Connors/The Lizard: Brilliantly-gifted herpetologist who lost most of his arm in a battlefield injury when he was in the US Armed Forces. His attempts to develop a means of regenerating lost limbs, canonically, led to the birth of his monstrous alter-ego.

Mary-Jane Watson: Girlfriend/Wife of Peter Parker, and often the one to provide the emotional support when he needs it.

Someone Else (Hero) There's plenty of others who can feature within this story setting
