Not all my family and friends make up my target audience - this has only very rarely created any issues. I write stories to entertain and while they are targeted at a certain audience, I like them to be available to a wide range of people.

As for critiquing, if one of them misses a theme or some other thing, I always look at it and decide if the person just missed it or if there is a problem with the writing. If all or most of them miss the theme or some other thing then I KNOW there's a problem with my delivery and I work on it.

Some of my friends and family make better critics than others but I welcome all their opinions. They however are not my only readers. I still rely on my writer's critique group too for a more detailed perspective.

Both are very important for me.
