Experimental Drug: This is a classic. The two main characters volunteer for an experimental drug study. The two will both be exposed to a drug and then kept in close proximity while they are under observation and being regularly subjected to examinations. The drug has a radically different reactions with testosterone vs estrogen. You can probably guess what these reactions will be.
Malfunction: Here we have a trip into space, to either a new planet, or a space station, or simply an extended test flight to test new systems in the crafts that will return them to earth. Due to some legal and bureaucratic details, men and women aren't allowed to fly together, so they will each fly in separate crafts (you can have it as each main character being alone in their respective crafts or have them each be part of a larger group all flying together). However, during the voyage, each craft suffers malfunctions, particularly with their simulated gravity. In the male vessel, the gravity is pretty much turned off and are left in an environment where exerting effort is almost impossible, while the female vessel has gravity greatly increased and it becomes a strain just to stand. These malfunctions happen rather early into the voyage, but aborting is not an option so they must continue onward and adapt to their environments. It's a long trip, so by the time they get off, the results are clearly shown.
Android: (I'll leave this option a little vague on purpose, there's a lot of different lines this could go down) You've basically got the idea of a male with a female android. The male might be the scientist who created her and is now testing and fine tuning his work. Or it could have gotten to the point where androids have become rather common and now the main character is just an average Joe with his android. Or somewhere in between. Either way, the android will be programmed to cater to the male's every need (possibly taking it too far) and while having a basic human female frame and scale, the components made it necessary to upscale things a bit so all the necessary parts would fit. So they solved this by taking a bit of artistic license and making the android in the image of a tall, muscled amazon. (Note that the android will probably be human-like in a Terminator fashion, you know, it'd be hard to just identify them as robots if you see them on the street, at least look human.)
A Study of Extremes: An interesting study is about to take place. A automated house has been built, one designed to take care of its inhabitants so there is no need to lift a finger (think Wall-e) and now a study is being proposed involving this. Two people will be placed in it. One is just an average Joe (maybe already a little on the heavy side) and the other is a female wilderness survival expert who has built a great deal of muscle from living out in the wilderness without any modern convenience. The male quickly becomes accustomed to being provided for and is soon engaging in little to no strenuous activity while the female, although no allowed to leave and go back to the wilderness, insists on a strict workout routine.
