Okay how about this: Spiderman tries to enhance his powers with more muscle with a spell that increases his muscle development & appetite; but it goes awry giving him fat instead, with a slowed down metabolism, & an exponentially stronger/faster-growing appetite than before.
Another option is that a new magical enemy (O.C.'s, any gender allowed or species of magical creatures from wizards, to demons, to nature spirits, etc.) is trying to fatten him up for some devious purpose like love/lust, food, strengthening themselves up as he gets fatter, etc.
Or what about the mystic half of his spider powers, born from a spider totem, is trying to tell him something, & the only way it communicates with him is during the night or when he feels his carnal desires acting up which will now be stimulated by lust/love, food, & his growing body.
